“Structures illustrate step-by-step mitochondrial transcription initiation”
转录起始是基因表达中的一个关键调控步骤,在这一过程中,RNA聚合酶(RNAP)从头开始合成RNA,并在特定长度的合成RNA触发过渡到延伸阶段。线粒体招募了一个单亚基RNAP和一个或两个辅助因子来启动转录。之前的研究已经揭示了酵母和人类线粒体RNAP起始复合物(ICs)的分子结构。
来自比利时吕汶大学的Kalyan Das和美国新泽西州皮斯卡塔韦市罗格斯大学的Smita S. Patel通过解析酵母线粒体RNAP和转录因子Mtf1的高分辨率冷冻电镜(cryo-EM)结构,提供了转录起始的全面、逐步机制,该机制在单核苷酸添加步骤中催化了2到8-核苷酸的RNA合成。不断增长的RNA-DNA在模板收缩和非模板重组的作用下适应于聚合酶缝隙中,形成了中间体。在早期起始阶段,非模板链的收缩和展开破坏了短的2到3-核苷酸的RNA合成,触发了合成失败。随后,非模板重新组织成一个基础堆叠的楼梯状结构,支持连续合成5到8-核苷酸的RNA。IC8中扩展的非模板阶梯和高度收缩的模板破坏了与Mtf1的启动子相互作用,从而促使启动子泡的破裂和启动子逃逸,完成了从起始到延伸复合物(EC)的过渡。一系列的转录起始步骤,每个步骤都由多个结构组分的相互作用引导,揭示了潜在的调控控制的精细调节机制。
